Biochemical High-Throughput Screening Assays for Inhibition of Tryptophan-Metabolizing Enzymes

Tryptophan metabolism plays an important role in immune modulation and neurodegeneration. Tryptophan-metabolizing enzymes are drug targets for cancer immunotherapy and neurodegenerative disease. We have developed NFK Green™ assays for the high-throughput screening of the tryptophan-metabolizing enzymes indoleamine 2,3-dioxygenase 1 (IDO1), indoleamine 2,3-dioxygenase 2 (IDO2), and tryptophan 2,3-dioxygenase (TDO).

Schematic representation of NFK Green™ assay. After enzymatic conversion of L-tryptophan into NFK, the reaction is stopped by addition of NFK Green™, which yields a fluorescent complex that can be detected at the indicated wavelengths.
